TRANSPORT Dr Tan Boon Keat * describe the quaternary structure of haemoglobin • To describe oxygen dissociation curve • To explain the factors that affect haemoglobin affinity to oxygen • To explain carbon dioxide transport * exclusively in the red blood cells (RBC) • main function: to transport O2 from the lungs to the capillaries of the tissues • consists of globin (protein) plus heme • belong to the group of proteins known as hemeproteins • Hemeproteins : a group of specialised proteins that contain heme as a tightly bound prosthetic group [other examples of hemeproteins: myoglobin, cytochromes & catalase ] * : globular protein consisting of four polypeptides  tetramer • Haemoglobin A : the major Hb in adults • composed of four polypeptide chains: 2  and 2 ß (2ß2)  quaternary structure • the chains are held together by non-covalent interactions • each polypeptide chain is a subunit * Lippincott's Illustrated Reviews: Biochemistry (7th Edition) Chapter Globular Protein * is composed of two identical dimers : (ß)1 and (ß)2 -Interchain hydrophobic interactions form strong associations between  and ß subunits in the dimer -Ionic and hydrogen bonds also occur between members of the dimer -The two dimers are able to move with respect to each other, being held together primarily by polar bonds * Lippincott's Illustrated Reviews: Biochemistry (7th Edition) Chapter Globular Protein * interactions between the mobile dimers result in the two dimers occupying different relative positions in deoxyhaemoglobin compared to oxyhaemoglobin • T (taut) form • the deoxy form of Hb • the two ß dimers interact through a network of ionic and hydrogen bonds that constraint the movement of the polypeptide chains • the low-oxygen affinity form of Hb * form: • the binding of O2 to Hb causes rupture of some of the ionic bonds and hydrogen bonds between the ß dimers • the polypeptide chains have more freedom of movement • the high oxygen-affinity form of Hb * 10.
